    Acreditação do Laboratório de Metrologia da Frilabo para Ensaios e Calibrações em Temperaturas

    O trabalho desenvolvido centrou-se na preparação da acreditação NP EN ISO/IEC 17025 do Laboratório de Metrologia da empresa Frilabo para prestação de serviços na área das temperaturas, no ensaio a câmaras térmicas e na calibração de termómetros industriais. Considerando o âmbito do trabalho desenvolvido, são abordados nesta tese conceitos teóricos sobre temperatura e incertezas bem como considerações técnicas de medição da temperatura e cálculo de incertezas. São também referidas considerações sobre os diferentes tipos de câmaras térmicas e termómetros. O texto apresenta os documentos elaborados pelo autor sobre os procedimentos de ensaio a câmaras térmicas e respetivo procedimento de cálculo da incerteza. Também estão presentes neste texto documentos elaborados pelo autor sobre os procedimentos de calibração de termómetros industriais e respetivo procedimento de cálculo da incerteza. Relativamente aos ensaios a câmara térmicas e calibração de termómetros o autor elaborou os fluxogramas sobre a metodologia da medição da temperatura nos ensaios, a metodologia de medição da temperatura nas calibrações, e respetivos cálculos de incertezas. Nos diferentes anexos estão apresentados vários documentos tais como o modelo de folha de cálculo para tratamento de dados relativos ao ensaio, modelo de folha de cálculo para tratamento de dados relativo às calibrações, modelo de relatório de ensaio, modelo de certificado de calibração, folhas de cálculo para gestão de clientes/equipamentos e numeração automática de relatórios de ensaio e certificados de calibração que cumprem os requisitos de gestão do laboratório. Ainda em anexo constam todas as figuras relativas à monitorização da temperatura nas câmara térmicas como também as figuras da disposição dos termómetros no interior das câmaras térmicas. Todas as figuras que aparecem ao longo do documento que não estão referenciadas são da adaptação ou elaboração própria do autor. A decisão de alargar o âmbito da acreditação do Laboratório de Metrologia da Frilabo para calibração de termómetros, prendeu-se com o facto de que sendo acreditado como laboratório de ensaios na área das temperaturas, a realização da rastreabilidade dos padrões de medida internamente, permitiria uma gestão de recursos otimizada e rentabilizada. A metodologia da preparação de todo o processo de acreditação do Laboratório de Metrologia da Frilabo, foi desenvolvida pelo autor e está expressa ao longo do texto da tese incluindo dados relevantes para a concretização da referida acreditação nos dois âmbitos. A avaliação de todo o trabalho desenvolvido será efetuada pelo o organismo designado IPAC (Instituto Português de Acreditação) que confere a acreditação em Portugal. Este organismo irá auditar a empresa com base nos procedimentos desenvolvidos e nos resultados obtidos, sendo destes o mais importante o Balanço da Melhor Incerteza (BMI) da medição também conhecido por Melhor Capacidade de Medição (MCM), quer para o ensaio às câmaras térmicas, quer para a calibração dos termómetros, permitindo desta forma complementar os serviços prestados aos clientes fidelizados à Frilabo. As câmaras térmicas e os termómetros industriais são equipamentos amplamente utilizados em diversos segmentos industriais, engenharia, medicina, ensino e também nas instituições de investigação, sendo um dos objetivos respetivamente, a simulação de condições específicas controladas e a medição de temperatura. Para entidades acreditadas, como os laboratórios, torna-se primordial que as medições realizadas com e nestes tipos de equipamentos ostentem confiabilidade metrológica1, uma vez que, resultados das medições inadequados podem levar a conclusões equivocadas sobre os testes realizados. Os resultados obtidos nos ensaios a câmaras térmicas e nas calibrações de termómetros, são considerados bons e aceitáveis, uma vez que as melhores incertezas obtidas, podem ser comparadas, através de consulta pública do Anexo Técnico do IPAC, com as incertezas de outros laboratórios acreditados em Portugal. Numa abordagem mais experimental, pode dizer-se que no ensaio a câmaras térmicas a obtenção de incertezas mais baixas ou mais altas depende maioritariamente do comportamento, características e estado de conservação das câmaras, tornando relevante o processo de estabilização da temperatura no interior das mesmas. A maioria das fontes de incerteza na calibração dos termómetros são obtidas pelas características e especificações do fabricante dos equipamentos, que se traduzem por uma contribuição com o mesmo peso para o cálculo da incerteza expandida (a exatidão de fabricante, as incertezas herdadas de certificados de calibração, da estabilidade e da uniformidade do meio térmico onde se efetuam as calibrações). Na calibração dos termómetros as incertezas mais baixas obtêm-se para termómetros de resoluções mais baixas. Verificou-se que os termómetros com resolução de 1ºC não detetavam as variações do banho térmico. Nos termómetros com resoluções inferiores, o peso da contribuição da dispersão de leituras no cálculo da incerteza, pode variar consoante as características do termómetro. Por exemplo os termómetros com resolução de 0,1ºC, apresentaram o maior peso na contribuição da componente da dispersão de leituras. Pode concluir-se que a acreditação de um laboratório é um processo que não é de todo fácil. Podem salientar-se aspetos que podem comprometer a acreditação, como por exemplo a má seleção do ou dos técnicos e equipamentos (má formação do técnico, equipamento que não seja por exemplo adequado à gama, mal calibrado, etc…) que vão efetuar as medições. Se não for bem feita, vai comprometer todo o processo nos passos seguintes. Deve haver também o envolvimento do todos os intervenientes do laboratório, o gestor da qualidade, o responsável técnico e os técnicos, só assim é que é possível chegar à qualidade pretendida e à melhoria contínua da acreditação do laboratório. Outro aspeto importante na preparação de uma acreditação de um laboratório é a pesquisa de documentação necessária e adequada para poder tomar decisões corretas na elaboração dos procedimentos conducentes à referida. O laboratório tem de mostrar/comprovar através de registos a sua competência. Finalmente pode dizer-se que competência é a palavra chave de uma acreditação, pois ela manifesta-se nas pessoas, equipamentos, métodos, instalações e outros aspetos da instituição a que pertence o laboratório sob acreditação.The developed work was focused on the accreditation NP EN ISO / IEC 17025 of the Frilabo Metrology Laboratory to provide services in the field of temperatures, in the thermal chambers test and calibration of industrial thermometers. Considering the scope of this work, are discussed in this thesis theoretical concepts of temperature and uncertainties as well as technical considerations of temperature measurement and calculation uncertainties. They are also referred to considerations about the different types of thermometers and thermal chambers. The text presents the documents prepared by the author on the procedures to test thermal chambers and respective uncertainty calculation procedure. They are also present in the texto documents drawned up by the author of the industrial thermometers calibration procedures and respective uncertainty calculation procedure. For the thermal chambers tests and thermometers calibration the author elaborated the flowcharts about temperature measurement methodology in testing, temperature measurement methodology in calibration, and respective calculations of uncertainty. The various annexes are presented several documents such as spreadsheet model for tests data analysis, spreadsheet model calibrations data analysis, model for test report template, model for calibration certificate template, spreadsheet models for customer/equipment management and automatic numbering of test reports and calibration certificates regarding to laboratory management requirements. Still attached there is all the figures relating to the temperature monitoring in the thermal chambers as well as the figures relating to thermometers disposition inside the thermal chambers. All figures that appear throughout the document that are not referenced are the adaptation or the author's own elaboration. The decision to extend the scope of accreditation of Frilabo Metrology Laboratory for thermometers calibration, caught up with the fact that being accredited as a testing laboratory in the field of temperature, achieving traceability of internal measurement standards, allow a optimized management resources. The methodology of the preparation of the entire accreditation process of the Frilabo Metrology Laboratory, was developed by the author and is expressed throughout the text of the thesis the relevant data for the realization of such accreditation in two areas. The evaluation of all the developed work will be carried out by the agency appointed IPAC (Portuguese Institute of Accreditation) which provides accreditation in Portugal. This accreditation organism will audit the company on the basis of developed procedures and the achieved results, and of these the most important to Balance the Best Uncertainty (BMI) measurement also known as Best Measurement Capability (MCM), or for testing the thermal chambers, or for the calibration of thermometers, thus allowing complement the services provided to loyal customers loyal to Frilabo. The thermal cameras and industrial thermometers are widely used in various industries, engineering, medicine, teaching and also research institutions, one of the goals respectively, the simulation of specific controlled conditions and temperature measurement. For accredited entities, such as laboratories, it is essential that the measurements taken with and in this types of equipment bearing metrological reliability, since inadequate results of measurements can lead to erroneous conclusions about the realised tests. The results obtained in thermal chambers test and thermometers calibration, are considered good and acceptable, since the best uncertainties obtained can be compared through public consultation of the IPAC Technical Annex, with the uncertainties of other accredited laboratories in Portugal. In a more experimental approach, we can say that in thermal chambers test to obtain lower or higher uncertainty mainly depends on the behavior, characteristics and chambers conservation state, concerning which the temperature stabilization process inside the same. Most sources of uncertainty in the thermometers calibration are obtained by the characteristics and the equipment manufacturer's specifications, which amount to a contribution of the same weight in calculating the expanded uncertainty (accuracy manufacturer, uncertainties inherited calibration certificates , stability and uniformity of the thermal environment where the calibrations are carried out). In thermometers calibration lower uncertainties are obtained for lower thermometers resolutions. It was found that the thermometers with a resolution of 1°C do not detected no variations in the thermal bath. In thermometers with lower resolutions, the weight of the contribution readings dispersion in the calculation of uncertainty, may vary depending on the thermometer characteristics. For example, thermometers with a resolution of 0.1°C, are the heaviest weight in the readings dispersion component of the contribution. It can be concluded that a laboratory accreditation is a process that is not at all easy. Aspects can be noted that may compromise the accreditation, such as poor selection in the technicians and equipment (untrained technician, equipment which is not suitable for example to the range, poorly calibrated, etc ...) that will perform the measurements. If not done well, it will compromise the whole process in the following steps. There must also be the involvement of all laboratory workers, quality manager, technical manager and technicians, only then is it possible to reach the required quality and continuous improvement of laboratory accreditation. Another important aspect in the preparation of a laboratory accreditation is the search of a necessary and proper documentation to be able to make correct decisions in the preparation of procedures leading to that. The laboratory has to show/prove by records its competence. Finally it can be said that competence is the keyword of an accreditation, as it manifests itself in people, equipment, methods, installations and other aspects of the institution to which it belongs under the laboratory accreditation

    Arbustus unedo essence: morphological and genetic characterization of the strawberry tree of Castelo de Paiva

    O medronheiro é um arbusto da região mediterrânica que pode ser encontrada por todo o país. Ao contrário do que verifica na região sul do país, no concelho de Castelo de Paiva é atribuída uma reduzida importância económica a esta espécie. Com o intuito de preservar e potenciar a produção desta espécie e contribuir para a dinamização da economia do concelho, procedeu-se à caracterização morfológica e genética de uma amostra da população de medronheiros de Castelo de Paiva. A caracterização morfológica e genética foi realizada para um total de 10 genótipos. Para tal recolheram-se 70 folhas aleatoriamente em cada árvore. Em 40 folhas mediu-se o comprimento, largura, comprimento do pedúnculo, peso fresco, peso seco e determinou-se a área foliar. Dos caracteres morfológicos analisados, aqueles que se revelaram mais úteis na distinção dos vários genótipos foram: comprimento do pedúnculo, peso fresco e peso seco. As restantes 30 folhas foram utilizadas para a caracterização genética. Esta caracterização foi realizada recorrendo a um marcador de DNA, ISSR. Os 5 primeiros exemplaresutilizados na técnica de ISSR demonstraram-se polimórficos. Os resultados da caracterização genética sugerem que a variabilidade genética na população é média a alta.The strawberry tree is a shrub native in the Mediterranean region and it can be found throughout Portugal. Unlike the case in the southern region of the country, in Castelo de Paiva a minor economic importance is given to this species. In order to preserve, to enhance the production of this species and to contribute to the boosting of the economy of the region, we proceeded to the characterization of a small sample population of this fruit tree of Castelo de Paiva in what concerns to its morphology and genetics. The morphological and genetic characterization was performed for a total of 10 genotypes. For this, 70 leaves were randomly collected from each tree. For 40 leaves, it was measured the length, the width, the peduncle length, the wet weight, the dry weight and determined the leaf area. Of the morphological characteristics analyzed, the ones that proved most useful in distinguishing the various genotypes were: the length peduncle, the wet weight and the dry weight. The remaining 30 leaves were used in the genetic characterization. This characterization was performed using a DNA marker, the ISSR. The 5 primers used in the ISSR technique proved to be polymorphic. The results from the genetic characterization suggest that variability in population genetics is medium to high

Assim o homem cresceu, dentro da sua mente, e a sua mente cresceu fora do homem: a linguagem sobrevive-nos, permite a transmissão de aquisições, veicula a cultura de geração em geração, está na base da civilização. A expansão humana consolida-se pelas técnicas de reprodução, mas com um preço: o da emancipação das coisas em relação aos homens. A linguagem pensa-nos. Os artefactos que produzimos exercem sobre nós estranhos poderes, as mercadorias são valorizadas tanto ou mais que as pessoas, o humano é desvalorizado na razão inversa.     SARS-CoV-2 introductions and early dynamics of the epidemic in Portugal

    Genomic surveillance of SARS-CoV-2 in Portugal was rapidly implemented by the National Institute of Health in the early stages of the COVID-19 epidemic, in collaboration with more than 50 laboratories distributed nationwide. Methods By applying recent phylodynamic models that allow integration of individual-based travel history, we reconstructed and characterized the spatio-temporal dynamics of SARSCoV-2 introductions and early dissemination in Portugal. Results We detected at least 277 independent SARS-CoV-2 introductions, mostly from European countries (namely the United Kingdom, Spain, France, Italy, and Switzerland), which were consistent with the countries with the highest connectivity with Portugal. Although most introductions were estimated to have occurred during early March 2020, it is likely that SARS-CoV-2 was silently circulating in Portugal throughout February, before the first cases were confirmed. Conclusions Here we conclude that the earlier implementation of measures could have minimized the number of introductions and subsequent virus expansion in Portugal.     The complete genome sequence of Chromobacterium violaceum reveals remarkable and exploitable bacterial adaptability

    Chromobacterium violaceum is one of millions of species of free-living microorganisms that populate the soil and water in the extant areas of tropical biodiversity around the world. Its complete genome sequence reveals (i) extensive alternative pathways for energy generation, (ii) ≈500 ORFs for transport-related proteins, (iii) complex and extensive systems for stress adaptation and motility, and (iv) wide-spread utilization of quorum sensing for control of inducible systems, all of which underpin the versatility and adaptability of the organism. The genome also contains extensive but incomplete arrays of ORFs coding for proteins associated with mammalian pathogenicity, possibly involved in the occasional but often fatal cases of human C. violaceum infection. There is, in addition, a series of previously unknown but important enzymes and secondary metabolites including paraquat-inducible proteins, drug and heavy-metal-resistance proteins, multiple chitinases, and proteins for the detoxification of xenobiotics that may have biotechnological applications

    As the knowledge base and importance of mitochondrial physiology to evolution, health and disease expands, the necessity for harmonizing the terminology concerning mitochondrial respiratory states and rates has become increasingly apparent. The chemiosmotic theory establishes the mechanism of energy transformation and coupling in oxidative phosphorylation. The unifying concept of the protonmotive force provides the framework for developing a consistent theoretical foundation of mitochondrial physiology and bioenergetics. We follow the latest SI guidelines and those of the International Union of Pure and Applied Chemistry (IUPAC) on terminology in physical chemistry, extended by considerations of open systems and thermodynamics of irreversible processes. The concept-driven constructive terminology incorporates the meaning of each quantity and aligns concepts and symbols with the nomenclature of classical bioenergetics. We endeavour to provide a balanced view of mitochondrial respiratory control and a critical discussion on reporting data of mitochondrial respiration in terms of metabolic flows and fluxes. Uniform standards for evaluation of respiratory states and rates will ultimately contribute to reproducibility between laboratories and thus support the development of data repositories of mitochondrial respiratory function in species, tissues, and cells. Clarity of concept and consistency of nomenclature facilitate effective transdisciplinary communication, education, and ultimately further discovery

    Cabbage and fermented vegetables : From death rate heterogeneity in countries to candidates for mitigation strategies of severe COVID-19

    Large differences in COVID-19 death rates exist between countries and between regions of the same country. Some very low death rate countries such as Eastern Asia, Central Europe, or the Balkans have a common feature of eating large quantities of fermented foods. Although biases exist when examining ecological studies, fermented vegetables or cabbage have been associated with low death rates in European countries. SARS-CoV-2 binds to its receptor, the angiotensin-converting enzyme 2 (ACE2). As a result of SARS-CoV-2 binding, ACE2 downregulation enhances the angiotensin II receptor type 1 (AT(1)R) axis associated with oxidative stress. This leads to insulin resistance as well as lung and endothelial damage, two severe outcomes of COVID-19. The nuclear factor (erythroid-derived 2)-like 2 (Nrf2) is the most potent antioxidant in humans and can block in particular the AT(1)R axis. Cabbage contains precursors of sulforaphane, the most active natural activator of Nrf2. Fermented vegetables contain many lactobacilli, which are also potent Nrf2 activators. Three examples are: kimchi in Korea, westernized foods, and the slum paradox. It is proposed that fermented cabbage is a proof-of-concept of dietary manipulations that may enhance Nrf2-associated antioxidant effects, helpful in mitigating COVID-19 severity.Peer reviewe

    Nrf2-interacting nutrients and COVID-19 : time for research to develop adaptation strategies

    There are large between- and within-country variations in COVID-19 death rates. Some very low death rate settings such as Eastern Asia, Central Europe, the Balkans and Africa have a common feature of eating large quantities of fermented foods whose intake is associated with the activation of the Nrf2 (Nuclear factor (erythroid-derived 2)-like 2) anti-oxidant transcription factor. There are many Nrf2-interacting nutrients (berberine, curcumin, epigallocatechin gallate, genistein, quercetin, resveratrol, sulforaphane) that all act similarly to reduce insulin resistance, endothelial damage, lung injury and cytokine storm. They also act on the same mechanisms (mTOR: Mammalian target of rapamycin, PPAR gamma:Peroxisome proliferator-activated receptor, NF kappa B: Nuclear factor kappa B, ERK: Extracellular signal-regulated kinases and eIF2 alpha:Elongation initiation factor 2 alpha). They may as a result be important in mitigating the severity of COVID-19, acting through the endoplasmic reticulum stress or ACE-Angiotensin-II-AT(1)R axis (AT(1)R) pathway. Many Nrf2-interacting nutrients are also interacting with TRPA1 and/or TRPV1. Interestingly, geographical areas with very low COVID-19 mortality are those with the lowest prevalence of obesity (Sub-Saharan Africa and Asia). It is tempting to propose that Nrf2-interacting foods and nutrients can re-balance insulin resistance and have a significant effect on COVID-19 severity. It is therefore possible that the intake of these foods may restore an optimal natural balance for the Nrf2 pathway and may be of interest in the mitigation of COVID-19 severity
